1. Needle insertion method: This method is suitable for large insects with hard body surface, and it is also the most commonly used specimen preparation method. After collection, before the specimen is dried, insert an insect needle on the specimen and do some work such as posture adjustment or spreading wings, and then finish it after drying.
2. Micro-needle and sticking method: For some small insects, ordinary insect needles are too big to be inserted into insects. At this time, it is necessary to insert the insect with a smaller microneedle, then insert the microneedle into a small cork, and then insert the ordinary insect needle into the cork. If there are smaller insects, you can stick the middle part of the right side of the insect to the tip of the small triangular paper with adhesive, and then insert the triangular paper with a general insect needle.
3. Soaking method: Some insects have soft body surfaces, such as termites or completely abnormal insects, and cannot be made into dry specimens for needle insertion. At this time, you can soak the worms in the liquid. Usually, insect tissues are fixed with special fixative or hot water first, and then soaked in 95% alcohol for preservation. Pay attention to the sealing of glass bottles, otherwise alcohol will easily volatilize. Sometimes the specimen will be dehydrated or the body fluid of the worm will flow out, which will reduce the alcohol concentration. At this time, alcohol must be changed several times.
4. Slide specimen: Slide specimen is suitable for tiny insects, and its morphological characteristics must be observed with microscope or magnifying glass. Such as Angel, fleas and aphids. The general steps are as follows: soak the collected specimens in 10% potassium hydroxide solution, soften the worm bones for one day, take them out, wash them with distilled water, and dye them with red dyes if necessary to facilitate observation. Then use the same concentration of 50- 100% alcohol for a series of dehydration, then seal with Arabic gum and dry for 2-3 weeks, and it will be a complete success.
